چکیده

The potato (Solanum tuberosum L.) cultivar 'SpuntaG2' contains a single copy of the Bacillus thuringiensis (Bt) cry1Ia1 gene and controls potato tuberworm (Phthorimaea operculella Zeller, Lepidoptera: Gelechiidae). Two potato cultivars and two breeding lines were crossed with SpuntaG2 creating four populations used to study cry1Ia1 segregation and efficacy. The cry1Ia1 gene segregated in each of the four populations with a 1:1 ratio. All progeny that were polymerase chain reaction positive for the cry1Ia1 gene had no surviving larvae and no leaf mining in detached leaf assays after 72 h. These results support previous evidence that SpuntaG2 carries a single copy of the cry1Ia1 gene and that transmission of the transgene from parent to progeny is not restricted and follows expected Mendelian segregation ratios. Based on detached leaf assays, the efficacy of the cry1Ia1 gene is retained through sexual transmission. If the SpuntaG2 cry1Ia1 insertion event is deregulated for commercial use, SpuntaG2 could be used for conventional breeding and the progeny carrying the SpuntaG2 event would also be available for commercial use.

Effects of Conunonly Used Insecticides on the Potato Tuherworm and Its Associated Parasites and Predators in Potatoes

J. Econ. Enlomol. 74: 30}-308 (1981) Larvae and adults of Phthorimaea operculella (Zeller) in potato foliage were effectively controlled by commercial applications of azinphosmethyl and a combination of methomyl and methamidophos, but eggs and pupae were not significantly affected. Azinphosmethyl, methomyl, and methamidophos provided adequate control of foliar larval populations, but larval par...

Postharvest Potato Tuherworm Population Levels in Cull and Volunteer Potatoes, and Means for Control

J. Eeon. Entomol. 73: 8-11 (1980) Postharvest samples of cull tubers and volunteer plants in commercial potato fields provided a large food resource for Phthorimaea opercu/ella (Zeller) during noncropping periods. In commercial, spring-planted fields sampled in 1977 and 1978, 27.6 and 74.3 surface culls/ 100 m2 and 1092.9 and 1670.0 subsurface culls/100 m2, respectively, were recovered 2 months...

Resistance of potato germplasm to the potato tuberworm (Lepidoptera: Gelechiidae).

The evaluation of potato germplasm for resistance to potato tuberworm, Phthorimaea operculella (Zeller) (Lepidoptera: Gelechiidae), is a valuable component of integrated pest management; however, few attempts have been made to identify natural genetic tuber resistance to tuberworm on potato germplasm. The objective of this study was to screen potato germplasm with potential tuberworm resistance...

Field and storage testing Bt potatoes for resistance to potato tuberworm (Lepidoptera: Gelichiidae).

Potato tuberworm, Phthorimaea operculella (Zeller), is the most serious insect pest of potatoes worldwide. The introduction of the Bacillus thuringiensis (Bt) toxin gene through genetic engineering offers host plant resistance for the management of potato tuberworm. We report on the field and storage studies to evaluate Bt-cry5 potato lines for resistance to potato tuberworm in Egypt under natu...

Performance of a North American Field Population and a Laboratory Colony of the Potato Tuberworm, Phthorimaea operculella, on Foliage of Resistant and Susceptible Potato Clones

Foliar resistance of two potato clones was tested against a Columbia Basin field population (CBFP) and a Colorado laboratory colony (COLC) of the potato tuberworm, Phthorimaea operculella (Zeller) (Lepidoptera: Gelechiidae). The first clone was a cross of a cultivated potato, Solanum tuberosum L. (Solanales: Solanaceae), and a wild potato, Solanum berthaultii Hawkes (Q 174-2); the second clone ...
